Overview of Synthetic Oat Avenanthramides

Hydroxyphenyl Propamidobenzoic Acid (HPPBA), also known as dihydroavenanthramide D, is a synthetic analogue of avenanthramides—naturally occurring compounds found in oats. Renowned for their soothing properties, avenanthramides have inspired the development of HPPBA to enhance skin comfort and health.

Skin Benefits of HPPBA

HPPBA offers a range of skin benefits, primarily due to its anti-inflammatory and anti-pruritic (anti-itch) properties.

Anti-Inflammatory Effects

HPPBA has been shown to reduce inflammation by inhibiting the release of pro-inflammatory cytokines. In fact, this action helps alleviate redness and swelling associated with various skin conditions. A study evaluating a non-steroidal cream containing HPPBA demonstrated significant reductions in inflammatory mediators, indicating its potent anti-inflammatory effects.1

Anti-Pruritic (Anti-Itch) Properties

In addition, HPPBA effectively diminishes itch sensations by modulating histamine release. In the same study, participants reported a notable decrease in pruritus, underscoring HPPBA's role in itch relief.1

Clinical Efficacy of HPPBA

The clinical efficacy of HPPBA has been substantiated through various studies.

In a clinical trial involving patients with seborrheic dermatitis, a cream containing HPPBA was applied twice daily over two weeks. Results showed a significant decrease in symptoms such as erythema, desquamation, and pruritus. Microbial cultures indicated a reduction in Malassezia furfur and Staphylococcus epidermidis, common contributors to skin irritation. Additionally, gene expression analysis revealed a downregulation of pro-inflammatory cytokines, confirming the anti-inflammatory and antimicrobial efficacy of HPPBA.1

These findings highlight HPPBA's potential as a therapeutic agent in managing inflammatory skin conditions, offering relief from discomfort and promoting overall skin health.

How is HPPBA Synthesised?

HPPBA is a lab-created compound inspired by avenanthramides, the skin-soothing molecules naturally found in oats. It is produced through a controlled synthetic process that replicates the beneficial structure of its natural counterpart. This repeatable and efficient method ensures high purity, stability, and efficacy.

The synthesis of HPPBA involves two key chemical reactions:2

  1. Building the Base Structure: Scientists start with p-hydroxyphenylpropionic acid, a molecule that mimics oat-derived avenanthramides. This compound is then chemically modified to enhance its stability and bioavailability in skincare formulations.
  2. Creating the Active Form: Next, the modified molecule undergoes a reaction with anthranilic acid—a naturally occurring compound—under carefully controlled conditions. This step forms the final bioactive structure, allowing HPPBA to deliver its anti-inflammatory and soothing benefits effectively when applied to the skin.

Synthesising HPPBA in a lab rather than extracting it from oats ensures consistency, purity, and long-term stability in skincare formulations. This method also allows for greater potency and better absorption, optimising HPPBA’s ability to calm irritation, reduce redness, and protect skin from environmental stressors.
